目的通过对人眼眶前脂肪细胞的培养和观察,了解眼眶前脂肪细胞的生物学特性,为进一步研究前脂肪细胞在一些眼部疾病如Graves眼病中的作用提供一种参考方法。方法用组织块培养法对人眼眶前脂肪组织进行体外培养,观察前脂肪细胞的形态变化及生长情况,绘制细胞生长曲线。以酶组织化学法检测细胞内甘油磷酸脱氢酶的含量。用油红O染色提取法测定细胞内脂肪含量。结果(1)在人眼眶前脂肪细胞的体外培养过程中,第4天可见前脂肪细胞自组织块爬出,第7天细胞内开始出现脂滴;(2)前脂肪细胞内甘油磷酸脱氢酶的含量自传代培养第3天开始上升,约第8天可达高峰;脂肪含量于第6天开始增加,约第11天达高峰;(3)人眼眶前脂肪细胞体外培养的倍增时间约为60 h。结论用组织块培养法可诱导人眼眶前脂肪细胞分化为成熟脂肪细胞。
ObjectiveTo culture the orbital preadipocyte of human and observe their biological characteristics,so as to provide a reference approach for further study on the role of the orbital preadipocytes in some eye diseases,e.g.Graves ophthalmopathy.MethodsTissue culture method was used in the cultivation of orbital adipose tissue in vitro.The morphological changes and growth of preadipocytes were observed, then the cell growth curve was recorded.The content of GPDH was determined by enzyme tissue chemical method.The intracellular fat accumulation was detected by Oil Red O staining.Results(1)During the in vitro cultivation of preadipocytes,the fat cells can be seen climbing out from the tissue on fourth day,and intracellular lipid droplets appeared on the seventh day.(2)The content of GPDH in preadipocytes began to increase on the 3rd day after subculture,about 8 days up to the peak.The content of fat began to increase on the sixth day,reached the peak on eleventh day approximately.(3)The doubling time of in vitro cultivation of human orbital preadipocyte is about 60 hours.ConclusionHuman orbital preadipocytes can be induced into mature fat cells by tissue cultivation method.
