目的建立大肠杆菌生物被膜(biofilm,BF)在腹膜透析留置管表面形成的体外模型,观察BF形成能力,比较在腹膜透析液及LB液环境中腹膜透析留置管大肠杆菌生物被膜形成的能力。方法采用腹膜透析液/LB液-腹膜透析留置管系统进行BF的培养,建立大肠杆菌BF体外模型,建模6 h和24 h,计算BF内活菌落数,银染法快速观察BF的变化,结晶染色法对载体表面生物膜进行半定量检测。结果腹膜透析留置管表面可形成大肠杆菌BF,6 h形成早期BF,24 h 形成成熟BF。不论是建模6 h还是24 h,LB液组BF内活菌数均高于腹透液组(P<0.01);银染后普通光学显微镜均可观察到黑染呈棉絮状的膜样物;结晶染色法LB液组BF半定量均高于腹透液组(P<0.01)。结论腹膜透析留置管表面可形成大肠杆菌生物被膜,LB液比腹膜透析液更易形成生物被膜。
ObjectiveTo establish in vitro model of escherichia coli biofilm,so as to observe and compare the forming ability of escherichia coli.MethodsBF was cultivated in peritoneal dialysis solution aimed to establish in vitro model of escherichia coli biofilm.After cultivated for 6 hours and 24 hours,BF was identified by rapid argentation. Bacterial colonies were calculated.ResultsWe could cultivate BF in the surface of detained tubes of peritoneal dialysis,forming early BF in 6 hours and forming mature BF in 24 hours.No matter cultivated for 6h or 24h,there were more bacterial colonies in LB liquid than in peritoneal dialysis solution(P<0.01).We could find black biofilm as wollige beschaffenheit through optical microscope.Crystal violet staining showed that LB liquid group′s semiquantitative values were higher than peritoneal dialysis solution group(P<0.01).ConclusionsEscherichia coli can establish BF on the surface of detained tubes of peritoneal dialysis.It is easier to form BF in the environment of LB liquid than peritoneal dialysis solution.
