甘草酸二铵对动物中枢胆碱酯酶和学习记忆力的影响

目的研究甘草酸二铵(diammonium glycyrrhizinate,DG)对家兔脑脊液中胆碱酯酶(acetylcholinesterase,AChE)活性的影响和对小鼠学习记忆力的影响。方法家兔耳缘静脉注射甘草酸二铵20~60 mg·kg-1·d-1;昆明种小鼠尾静脉注射甘草酸二铵20~60 mg·kg-1·d-1。两周后,采脑脊液测定家兔脑脊液胆碱酯酶活性,通过Y形迷宫实验观察对小鼠学习记忆的影响。结果应用甘草酸二铵组的家兔脑脊液中胆碱酯酶活性明显低于生理盐水组(P<0.05);应用甘草酸二铵高剂量组的小鼠获得记忆所需电击次数少于生理盐水组(P<0.05)。结论甘草酸二铵可抑制脑脊液胆碱酯酶活性,增强学习记忆力。
ObjectiveTo investigate the effect of diaminonium glycyrrhizinate (DG) on the activity of acetylcholinesterase (AChE) in cerebrospinal fluid of rabbits and the learning and memory ability of mice.MethodsThe injection of DG was performed on mice(20-60mg/kg iv)and rabbits (20-60 mg/kg iv) once everyday.After 2 weeks,the activity of AChE in cerebrospinal fluid of rabbits was determined.Y-maze test was designed to study the learning and memory ability of mice.Resultsthe activity of AChE of the group injected with DG was lower than that of the group injected with sodium chloride(P<0.05);The number of electric shocks of the group injected with DG(high dosage)significantly decreased in Y-maze(P<0.05).ConclusionThe DG has an inhibitory effect on the activity of AChE in cerebrospinal fluid and has the effects of improving learning and memory.
