目的探讨HIV-1感染过程中调节性T细胞(Treg)上PD-L1的表达水平及其与外周血Treg频率的相关性。方法采集36例未经抗病毒治疗的HIV-1感染者及16例健康对照者外周血,提取单个单核细胞(PBMCs),用流式细胞技术检测Treg上PD-L1的表达情况,计算Treg频率。结果HIV-1感染组患者Treg PD-L1表达率显著低于对照组健康志愿者,差异具有统计学意义(P<0.01)。HIV-1感染者Treg上PD-L1的表达百分比与外周血CD3+CD4+T细胞中Treg的频率呈正相关(r=0.6305,P<0.01)。结论HIV-1感染过程中,Treg上PD-Ll表达升高,可能与Treg频率的升高有关。
ObjectiveTo investigate the expression of PD-L1 on Treg cells and the correlation with the frequency of peripheral blood Treg in patients with HIV-Ⅰ. MethodsPeripheral blood from 36 patients with HIV-Ⅰ(patients without antiretroviral therapy, HIV-Ⅰinfection group) and 16 healthy persons ( control group ) was collected, PBMCs were abstracted, flow cytometry was used to test the expression of PD-L1 on regulatory T cells(Treg) and calculate the Treg frequency. ResultsThe expression of PD-L1 on Treg cells in the HIV-Ⅰinfection group was significantly lower than that in the control group (P<0.01), that was a positive correlation between the expression of PD-L1 on Treg cells and Treg frequency on peripheral blood T cells of CD3+, CD4+ in patients with HIV-Ⅰ(r=0.6305,P<0.01). ConclusionsThe up-regulated expression of PD-L1on Treg may be associated with the increased frequency of Treg in patients with HIV-Ⅰ.
