目的探讨胸腺五肽(thymopentin,TP-5)对大鼠腹腔巨噬细胞吞噬功能的影响。方法取13只大鼠,均给予腹腔注射5%淀粉溶液刺激其产生并激活巨噬细胞,72 h后灌洗提取巨噬细胞。制备金黄色葡萄球菌悬液,并分为两组,实验组加入5 μL胸腺五肽(浓度:1 mg/mL),对照组加入等体积的生理盐水,每组均设13个亚组,孵育30 min后,每个亚组均加入450 μL巨噬细胞悬液。孵育40 min后进行细胞涂片,吉姆萨染色后于油镜下观察细胞形态并计算巨噬细胞的平均吞噬率和平均吞噬细菌个数。结果实验组吞噬细菌的巨噬细胞数、巨噬细胞吞噬细菌总数以及巨噬细胞的平均吞噬率、平均吞噬细菌个数均显著多于或高于对照组(P<0.01)。结论胸腺五肽可能是通过改变金黄色葡萄球菌结构及(或)增强巨噬细胞作用,从而提高大鼠腹腔巨噬细胞吞噬细菌的功能。
ObjectiveTo investigate the effects of thymopentin on macrophages phagocytosis of enterocoelia in rats. MethodsA total of 13 Wistar rats were injected intraperitoneal with 5% starch solution to generating macrophages.Then after 72 hours of injection, enterocoelia macrophages were lavaged and fetched. Staphylococcus aureus suspension was divided into two groups, the experimental group was mixed with 5 μL TP-5(effective concentration:1mg/mL),while the control group was mixed with the same volume of saline, and 13 subgroups were set in each group, 450 μL macrophage suspension was added to each subgroup after 30 minutes of incubation. Cell smear was conformed after 40 minutes of incubation in vitro, Giemsa staining was used to detect the cell smear and cellular morphology was observed on the glass slides, the phagocytosis rate and phagocytosis number were calculated. ResultsThe number of macrophages which phagocytosised bacteria, the total number of bacteria which phagocytosised by macrophages, the average rate of phagocytosis and the average of phagocytosis number in experimental group were significantly more or higher than those of the control group (P<0.01). ConclusionsThymopentin may be can change the structure of Staphylococcus aureus and (or) enhance the macrophage cytotoxicity,which could improve the function of macrophages to swallow Staphylococcus aureus.
