目的初步建立冠心病患者与健康对照的血小板蛋白质表达图谱,并鉴定其差异表达的蛋白质。方法选取冠心病患者9例和健康志愿者11名作为研究对象,抽取外周血,分离血小板并提取蛋白质。采用二维凝胶电泳技术分离并建立血小板蛋白质表达图谱,以ImageMaster 2D platinum 5.0软件进行图谱分析,选取差异点进行基质辅助激光解析/电离-飞行时间质谱(Matrix-Assisted Laser Desorption/ Ionization Time of Flight Mass Spectrometry,MALDI-TOF-TOF)分析,再采用Mascot软件搜索数据库鉴定差异表达蛋白质。结果(1)初步建立了两组对象的血小板蛋白质表达图谱;(2)筛选出17个差异蛋白点,质谱成功鉴定7个:Plastin-2、Coronin-1A、Clusterin、 Annexin A1、 Chloride intracellular channel protein 1、Peroxiredoxin-6、Peroxiredoxin-2。结论血小板蛋白Plastin-2、Coronin-1A、Clusterin、 Annexin A1、Peroxiredoxin-6、Peroxiredoxin-2在冠心病的发生发展中可能起到关键作用。
ObjectiveTo establish the protein expression profile of platelet in patients with coronary heart disease and healthy control, and identify the differentially expressed proteins. MethodsNine patients with coronary heart disease and eleven healthy volunteers were enrolled. Peripheral blood of all subjects was extracted, platelet was isolated and protein was extracted. Two dimensional gel electrophoresis technique was used to isolate and establish the protein expression profile of platelet. ImageMaster 2D platinum 5.0 software was used to analyze the spectrum, differential expression protein was selected and identified by MALDI-TOF-TOF and Mascot software,respectively. Results(1) Preliminary establisht of Platelet protein expression profiles of the two groups was preliminary established; (2) 17 differentially expressed protein spots were identified, and 7 were successfully identified by mass spectrometry: Plastin-2,Coronin-1A,Clusterin,Annexin A1,Chloride intracellular channel protein 1、Peroxiredoxin-6、Peroxiredoxin-2. ConclusionsThe platelet protein, including Plastin-2,Coronin-1A,Clusterin,Annexin A,Peroxiredoxin-6,Peroxiredoxin-2 may play a key role in the occurrence and development of coronary heart disease.
