目的探讨柔红霉素(Daunorubicin, DNR)对急性早幼粒细胞白血病(APL)细胞株HL-60细胞中细胞因子信号转导抑制蛋白1(SOCS1)-mRNA、细胞因子信号转导抑制蛋白3(SOCS3)-mRNA表达的影响。方法取HL-60细胞株,设HL-60组、HL-60+DNR组,同时取3例正常人外周血白细胞设为NC组。NC及HL-60组未给予干预,HL-60+DNR组为小剂量DNR持续作用HL-60细胞10 d。提取总RNA进行,采用实时荧光定量聚合酶链反应(RT-PCR)法检测SOCS1-mRNA、SOCS3-mRNA转录水平,每组实验重复3次。结果与NC组相比,HL-60组及HL-60+DNR组SOCS1-mRNA转录水平均显著下调(P<0.05),HL-60组下调更明显(P<0.05)。与NC组相比,HL-60组SOCS3-mRNA转录水平显著上调(P<0.05),而HL-60+DNR组上调不明显(P>0.05)。结论DNR可能通过上调HL-60细胞珠SOCS1及下调其SOCS3的表达,诱导APL细胞的凋亡及促进其成熟,促进急性早幼粒细胞白血病患者病情缓解。
ObjectiveTo explore the effect of daunorubicin(DNR) on the expression of suppressor of cytokine signaling 1 (SOCS 1) -mRAN and SOCS 3 -mRNA in acute promyelocytic leukemia cell line HL-60 cells. MethodsHL-60 cell line was set to HL-60 group, HL-60+DNR group, and peripheral blood leukocytes were separated from the blood of 3 normal subjects as NC group. The HL-60 group and NC group received no treatment, while the HL-60+DNR group was exposed to small doses of DNR every two day for 10 days. Total RNA of all groups was extracted and SOCS1-mRNA and SOCS3-mRNA were detected by real-time reverse transcription polymerase chain reaction(RT-PCR). The analysis for each group was performed for three times. ResultsThe SOCS1-mRNA expression in the HL-60 groups and HL-60+DNR group were markedly down-regulated compared to the NC group(P<0.05), and it was more markedly in HL-60 group(P<0.05). The SOCS3-mRNA expression in the HL-60 groups was markedly up-regulated compared to the NC group(P<0.05), and it was up-regulated but not obviously in the HL-60+DNR group(P>0.05). ConclusionsDNR can up-regulate the expression of SOCS1 and down-regulate the expression of SOCS3 in HL-60 cell line, which may induce the apoptosis and maturation of leukemia cells and promote remission in APL patients.
