目的探讨高脂饮食联合维生素D3腹腔注射对大鼠动脉组织AU碱基富集区结合因子1(AUF1) 及白细胞介素35(IL-35)表达的影响。方法选择SD大鼠20只随机分为实验组和对照组各10只。给予实验组大鼠高脂饮食及维生素D3腹腔注射,对照组大鼠予以正常饲养。12周后检测比较两组大鼠的血脂水平、动脉组织病变情况;比较两组大鼠动脉组织AUF1 mRNA及IL-35 mRNA及其蛋白表达情况。结果实验组大鼠血清总胆固醇、三酰甘油均显著高于对照组,动脉组织AUF1 mRNA、IL-35 mRNA表达显著高于对照组(P<0.05)。光镜下,对照组大鼠动脉内膜光滑,中膜平滑肌细胞排列整齐,纤维清晰,边界清楚,无炎症细胞浸润;实验组大鼠主动脉内皮细胞肿胀增厚,中膜平滑肌细胞排列紊乱,局部有轻微增殖等表现,但未见斑块组织。两组大鼠动脉组织AUF1及IL-35的蛋白印迹均未见明显条带。结论高脂饮食联合维生素D腹腔注射能诱发大鼠动脉发生一定程度的炎症反应,对动脉组织的AUF1 mRNA、IL-35 mRNA表达产生比较明显的影响,但对动脉组织IL-35及AUF1蛋白表达并未产生明显的影响,可能与本研究大鼠实验周期较短有关。AUF1、IL-35可作为抗炎症性反应干预的靶点。
ObjectiveTo investigate the effects of high-fat diet combined with vitamin intraperitoneal injection D3 on the expression of AU-rich element RNA-Binding factor 1 (AUF1) and interleukin 35 (IL-35) in rat arterial tissue. Methods20 SD rats were randomly divided into experimental group and control group. The experimental group were given a high-fat diet and intraperitoneal injection of vitamin D3. The control group was raised normally. After 12 weeks, the blood lipid levels and arterial tissue lesions were compared between the two groups. The mRNA and protein expressions of AUF1 and IL-35 were compared between the two groups. ResultsThe serum total cholesterol and triglyceride in the experimental group were significantly higher than those in the control group, whose the expression of AUF1 mRNA and IL-35 mRNA in the arterial tissue was significantly higher than that in the control group (P<0.05). Under light microscope, the arterial intima of the control group was smooth, the smooth muscle cells of the middle membrane were arranged neatly, the fibers were clear, the boundary was clear, and there was no infiltration of inflammatory cells. In the experimental group, the aortic endothelial cells of the experimental group were swollen and thick, and the smooth muscle cells of the medium were disordered. There was a slight local proliferation, but no plaque tissue. There were no obvious bands in the protein blots of AUF1 and IL-35 in the arteries of the two groups. ConclusionThe high-fat diet combined with intraperitoneal injection of vitamin D can induce a certain degree of inflammatory reaction in rat arteries, which has a significant effect on the expression of AUF1 mRNA and IL-35 mRNA in arterial tissue. However, it did not have a significant effect on the expression of IL-35 and AUF1 protein in arterial tissue, which may be related to the shorter experimental period of the rats in this study. AUF1 and IL-35 can be used as targets for anti-inflammatory response.
