目的探讨外周血hsa_circRNA_104600的表达水平与急性缺血性脑卒中的关系。方法选取2017年8月至2019年9月在广西中医药大学第一附属医院神经内科住院治疗的急性缺血性脑卒中患者45例作为病例组,根据《美国国立卫生研究院卒中量表》(NIHSS)评分分为轻度神经功能缺损组(<5分,32例)、中重度神经功能缺损组(≥5分,13例);选取同期在南宁市青秀区2家社区卫生服务中心参加健康体检的健康志愿者45人作为对照组。检测比较急性缺血性脑卒中患者和对照组健康志愿者外周血hsa_circRNA_104600的表达水平;绘制根据hsa_circRNA_104600水平诊断急性缺血性脑卒中的ROC曲线;比较轻度神经功能缺损组与中重度神经功能缺损组急性缺血性脑卒中患者外周血的hsa_circRNA_104600表达水平。结果病例组急性缺血性脑卒中患者的外周血hsa_circRNA_104600表达水平显著高于对照组健康志愿者(P<0.05)。由ROC曲线得出,根据hsa_circRNA_104600外周血表达水平诊断急性缺血性脑卒中的ROC曲线下面积(AUC)为0.725、灵敏度为0.933、特异度为0.533。中重度神经功能缺损组急性缺血性脑卒中患者的外周血hsa_circRNA_104600表达水平显著高于轻度神经功能缺损组患者,差异有统计学意义(P<0.05)。结论外周血hsa_circRNA_104600表达水平与急性缺血性脑卒中患者的发病及其神经功能缺损的严重程度存在比较密切的关系,可作为潜在的治疗靶点。
ObjectiveTo explore the relationship between the expression level of hsa_circRNA_104600 in peripheral blood and acute ischemic stroke. MethodA total of 45 patients with acute ischemic stroke who were hospitalized in the Neurology Department of the First Affiliated Hospital of Guangxi University of Traditional Chinese Medicine from August 2017 to September 2019 were selected as the case group, and they were divided into mild neurological deficit group (<5 points, 32 cases) and moderate to severe neurological deficit group (≥5 points, 13 cases) according to the National Institutes of Health Stroke Scale (NIHSS) scores; meanwhile, 45 healthy volunteers who participated in physical examinations in 2 Community Health Service Centers in Qingxiu District, Nanning during the same period were selected as the control group. The expression level of hsa_circRNA_104600 in peripheral blood of patients with acute ischemic stroke and healthy volunteers in the control group was detected and compared. The ROC curve was drawn based on hsa_circRNA_104600 level in the diagnosis of acute ischemic stroke. The expression level of hsa_circRNA_104600 in the peripheral blood of patients with acute ischemic stroke in the mild neurological deficit group and the moderate to severe neurological deficit group was compared. ResultsThe expression level of hsa_circRNA_104600 in peripheral blood of patients with acute ischemic stroke in the case group was significantly higher than that of healthy volunteers in the control group (P<0.05). The ROC curve results showed that the area under the ROC curve (AUC) based on the expression level of hsa_circRNA_104600 in peripheral blood for the diagnosis of acute ischemic stroke was 0.725, the sensitivity was 0.933, and the specificity was 0.533. The expression level of hsa_circRNA_104600 in peripheral blood of patients with acute ischemic stroke in the moderate to severe neurological deficit group was significantly higher than that of patients in the mild neurological deficit group, and the difference was statistically significant (P<0.05). ConclusionThe expression level of hsa_circRNA_104600 in peripheral blood is closely related to the onset of acute ischemic stroke and the severity of neurological deficits, which can be used as a potential therapeutic target.
