E2F5在肝细胞肝癌中的表达及其与患者预后关系的研究

目的探讨E2F5在肝细胞肝癌(Hepatocellular carcinoma,HCC)组织中的表达水平;分析E2F5的表达水平与HCC患者的性别、年龄、血清AFP及ki-67浓度、肿瘤病理学分级、肿瘤TNM分期及患者预后的关系。方法通过癌症基因组图谱(TCGA)数据库获取HCC患者的E2F5基因组学数据、临床资料及预后信息,比较HCC患者癌组织和癌旁正常组织中的E2F5表达水平;比较E2F5低表达组和E2F5高表达组HCC患者的性别、年龄、肿瘤病理学分级、肿瘤TNM分期、血清AFP水平、ki-67水平、患者的整体生存率。结果HCC患者癌组织的E2F5表达水平显著高于其癌旁正常组织的表达水平,差异有统计学意义(P<0.05)。E2F5高表达组HCC患者的病理学分级、肿瘤TNM分期、血清AFP水平、血清ki-67水平均显著高于E2F5低表达组患者,差异有统计学意义(P<0.05),但患者的性别、年龄比较,差异无统计学意义(P>0.05)。Kaplan-Meier生存曲线显示,E2F5高表达组HCC患者的整体生存率显著低于E2F5低表达组患者,差异有统计学意义(P<0.05)。结论HCC患者癌组织中的E2F5呈高表达,E2F5高表达患者的肿瘤病理学分级及TNM分期、血清AFP水平、血清ki-67水平均显著高于E2F5低表达患者,HCC患者E2F5的高表达与其不良预后密切相关。
ObjectiveTo investigate the expression of E2F5 in hepatocellular carcinoma (HCC) tissues, and to analyze the relationship between E2F5 expression level and the gender, age, the concentration of serum AFP and ki-67, tumor pathological grade, TNM stage, as well as prognosis of the patients. MethodsE2F5 genomics data, clinical data and prognostic information of HCC patients were obtained from The Cancer Genome Atlas (TCGA) database. The expression levels of E2F5 between the cancer tissues and normal tissues adjacent to cancer of HCC patients were compared. The gender, age, tumor pathological grade, TNM stage, serum AFP level and ki-67 level, the overall survival rate of HCC patients between the E2F5 low expression group and the E2F5 high expression group were compared. ResultsThe expression level of E2F5 in cancer tissues of HCC patients was significantly higher than that in normal tissues adjacent to cancer, with a statistically significant difference (P<0.05). The pathological grade, TNM stage, serum AFP level, and serum ki-67 level of HCC patients in the E2F5 high expression group were significantly higher than those in the E2F5 low expression group, with statistically significant differences (P<0.05), but there were no statistically significant differences in gender and age between the two groups (P>0.05). The Kaplan-Meier survival curve showed that the overall survival rate of HCC patients in the E2F5 high expression group was significantly lower than that in the E2F5 low expression group, with a statistically significant difference (P<0.05). ConclusionE2F5 is highly expressed in the cancer tissues of HCC patients. The tumor pathological grade and TNM stage, serum AFP level, and serum ki-67 level of HCC patients with high E2F5 expression are significantly higher than those of patients with low E2F5 expression. The high expression of E2F5 in HCC patients is closely related to its poor prognosis.
