目的探究不同浓度岩黄连总碱(CSBTA)对牙龈卟啉单胞菌(P.gingivalis)体外生长的影响。方法检测CSBTA对P.gingivalis的最小抑菌浓度(MIC)和最小杀菌浓度(MBC)。用不同浓度(0.01 g/L、0.02 g/L、0.04 g/L、0.08 g/L、0.16 g/L、0.32 g/L)CSBTA溶液对P.gingivalis进行处理,检测其对P.gingivalis的相对生长抑制率;用1/4MIC、1/2MIC、MIC的CSBTA溶液对P.gingivalis进行处理,观察并检测其对P.gingivalis生长的影响,并绘制生长曲线。结果CSBTA对P.gingivalis的MIC为0.08 g/L,MBC为0.16 g/L。CSBTA浓度为0.02 g/L时,50%以上细菌生长受到抑制[相对生长抑制率为(55.36±7.39)%];CSBTA浓度为0.08 g/L时,90%以上细菌生长受到抑制[相对生长抑制率为(93.21±3.38)%]。0.02 g/L、0.04、0.08 g/L CSBTA溶液对P.gingivalis生长均有不同程度的抑制,且呈浓度依赖性趋势,0.02 g/L、0.04 g/L CSBTA溶液处理时P.gingivalis缓慢生长,而0.08 g/L CSBTA溶液处理时P.gingivalis生长几乎完全受到抑制。结论CSBTA对P.gingivalis的体外生长具有较强的抑制作用。
ObjectiveTo explore the effects of different concentrations of Corydalis saxicola Bunting total alkaloids (CSBTA) on the Porphyromonas gingivalis(P.gingivalis) in vitro growth. MethodsThe minimum inhibitory concentration(MIC) and the minimum bactericidal concentration(MBC) of CSBTA against P.gingivalis were determined. P.gingivalis was treated with different concentrations(0.01 g/L, 0.02 g/L, 0.04 g/L, 0.08 g/L, 0.16 g/L, 0.32 g/L) of CSBTA solutions to detect their relative growth inhibitory rates against P.gingivalis. P.gingivalis was treated by 1/4 MIC, 1/2 MIC, and MIC of CSBTA solutions to observe and detect their impacts on the growth of P.gingivalis, and the growth curve was drawn.ResultsThe MIC of CSBTA against P.gingivalis was 0.08 g/L, and the MBC was 0.16 g/L. It inhibited over 50% of bacteria′s growth when the concentration of CSBTA reached 0.02 g/L(the relative growth inhibitory rate was [55.36±7.39]%) and over 90% of bacteria′s growth when the concentration of CSBTA reached 0.08 g/L (the relative growth inhibitory rate was [93.21±3.38]%). The 0.02 g/L, 0.04 g/L, and 0.08 g/L CSBTA solutions inhibited the growth of P.gingivalis to varying degrees, and the inhibition showed concentration dependence. Therefore,P.gingivalis grew slowly when treated with the 0.02 g/L or 0.04 g/L CSBTA solution, while its′ growth was almost completely inhibited when treated with the 0.08 g/L CSBTA solution. ConclusionCSBTA had a relevantly strong inhibitory effect on the growth of P.gingivalis in vitro.
