精索静脉曲张程度对精子DNA完整性和α-1,4-葡糖苷酶含量的影响

目的研究精索静脉曲张程度对精子DNA完整性和精液中α-1,4-葡糖苷酶含量的影响。方法选取就诊于广西医科大学第一附属医院生殖医学研究中心、男性科门诊并确诊为精索静脉曲张患者84例,并按精索静脉曲张程度分为3组,B组:1级精索静脉曲张,共26例;C组:2级精索静脉曲张,共28例;D组:3级精索静脉曲张,共30例;另外选取27例确认有生育能力的健康男性的精液样本作为对照组(A组)。采用单向凝胶电泳技术(SCGE)对精子DNA完整性进行分析,硝基酚法检测精液中α-1,4-葡糖苷酶含量。结果(1)3组精索静脉曲张患者的精子DNA完整性和精液中α-1,4-葡糖苷酶含量均低于对照组,差异有统计学意义(P<0.05)。(2)1级和2级,3级精索静脉曲张患者的精子DNA完整性比较,差异有统计学意义(P<0.05),2级和3级精索静脉曲张患者的精子DNA完整性无统计学差异(P>0.05)。(3)1级和2级,3级精索静脉曲张患者精液中的α-1,4-葡糖苷酶含量比较,差异有统计学意义(P<0.05),2级和3级精索静脉曲张患者精液中的α-1,4-葡糖苷酶含量差异有统计学意义(P<0.05)。结论精索静脉曲张可导致男性患者精子DNA完整性下降及精液中的α-1,4-葡糖苷酶含量降低,进而影响到精子质量,且精索静脉曲张程度越重,影响越大。
ObjectiveTo investigate the integrity of sperm nuclear DNA and the content ofα-1, 4-glucosidase in patients with varicocele of different degrees. MethodsThe semem samples from 84confirmed patients with varicocele from the department of andriatrics and reproductive centre of the First Affiliated Hospital of Guangxi Medical University were assigned into 3 groups(B,C,D) based on varicocele degree. The samples of group B,C,D contained 26,28,30 cases respectively. 27 semem samples from normal fertile men were collected as control group (group A). Single cell gel electropherosis (SCGE)was applied to assess the integrity of sperm nuclear DNA. The content of α-1, 4-glucosidase were detected by Nitrophenol method. Results(1) The integrity of sperm nuclear DNA and the content of α-1, 4-glucosidase in patients with varicocele were significantly lower than those in the control group (P<0.05). (2) The integrity of sperm nuclear DNA in group B was significantly higher than that in the group C and group D (P<0.05). However, there was no significant difference in sperm nuclear DNA integrity between group C and group D. (3)The content of α-1, 4-glucosidase in group B was significantly higher than that in the group C and group D (P<0.05), and the content of α-1, 4-glucosidase in group C was also significantly higher than that in the group D (P<0.05). ConclusionsVaricocele might lead to the decrease of sperm DNA integrity and the content of α1, 4-glucoside, and affect sperm quality, the more severe of varicocele, the greater the impact.
